The fiber optic fluorescence flow cell cell configuration (A) can be either placed within LOV module or attached externally (B). The advantages of the external configuration are that the flow call can be thermostated, it can be fabricated from opaque material, to minimize external illumination and that the volume of the channel between the central port and light path can be changed by selecting a suitable insert. The insert of orange PEEK tubing shown here forms a channel with 10 microliter volume.
The calibration run was obtained using Ocean Optics Fluorometer , Tungsten lamp light source and software protocol (C) using DI water as carrier and fluorescein standards in an ammonium acetate buffer pH 7.5.
